Clue emerged from the enforced leisure of wartime Britain. Anthony E. Pratt conceived the murder mystery game during World War II air-raid blackouts, designing a deduction puzzle set in a Victorian mansion where players collect evidence to identify the murderer, weapon, and location. His wife assisted in developing the playing board. The game's name—Cluedo in Britain—combined the word "clue" with "ludo," the Latin term meaning "I play," connecting it to the ancient Indian game of Pachisi. When Pratt sold the patent to waddingtons.com and its American partner parkerbrothers.com, the American publisher renamed it simply Clue, as ludo held no cultural currency in the United States.

The original design featured six suspect characters, six weapons, and nine rooms within the mansion. Pratt's patent had included ten characters and additional implements such as a shillelagh and hypodermic syringe, but the commercial version was streamlined for manufacturing efficiency. Early tokens were crafted from period materials: the rope was actual string, and the lead pipe was fashioned from genuine lead. As manufacturing standards and safety awareness evolved, these were replaced with plastic and then pewter alternatives. The 2023 edition introduced character backstories—Mrs. White as a chef, Mrs. Peacock as a lawyer—and replaced generic colored pawns with sculpted metal character movers.
